Zusammenfassung
Pentacyclische Triterpene stellen aufgrund ihres starren Grundgerüsts eine strukturell anspruchsvolle Naturstoffklasse dar. Ziel dieser Arbeit war die gezielte chemische Modifikation von Betulin, Betulinsäure, Oleanol- und Ursolsäure zur Entwicklung neuer KRas4B-Inhibitoren. Durch Acylierungen, Amidkupplungen, Oxidationen und die Einführung von Phthalat-, Hydroxamsäure- und Tetrazolgruppen wurde eine Bibliothek funktionalisierter Derivate synthetisiert. Besonders Phthalat-Substituenten in Position 3 führten zu einer deutlichen Steigerung der Aktivität im GDP/GTP-Austauschassay (IC50-Werte bis in den nanomolaren Bereich gegen KRasG12D und im einstellig mikromolaren Bereich gegen KRasG12V). Glide-Docking- und Moleküldynamikstudien identifizierten eine bislang unbeschriebene allosterische Bindetasche an dem KRas4Ballo:SOS-Interface, in der die Dimethoxyphthalat-Derivate stabile Komplexe (Dockingscores bis -12.2 kcal/mol) bilden. Das zentrale Bindungsmotiv beschreibt die Chelatisierung des Mg2+-Ions durch einen Phthalat-Rest, während sich das Grundgerüst entlang der Rasallo-SOSRem-Interface anlagert. Dadurch wird die Tyr64Ras-SOSRem-Interaktion gestört, die für die Stabilisierung der aktiven SOS-Konformation verantwortlich ist. Durch die Störung dieser Wechselwirkung kann der GDP/GTP-Austausch nicht mehr stattfinden. Trotz begrenzter „drug-likeness“ markieren phthalatmodifizierte Triterpene einen vielversprechenden Ausgangspunkt für die Entwicklung strukturell distinkter, allosterischer Pan-Ras-Inhibitoren, die auch die schwer adressierbare KRasG12V-Mutante erfassen.
Abstract
Pentacyclic triterpenes represent a structurally demanding class of natural products due to their rigid polycyclic carbon framework. The aim of this study was the targeted chemical modification of betulin, betulinic acid, oleanolic acid, and ursolic acid to develop novel KRas4B inhibitors. A library of functionalized derivatives was synthesized by employing acylations, amide couplings, oxidations, and the introduction of phthalate, hydroxamic acid, and tetrazole moieties. Phthalate substituents in position 3 proved particularly effective, leading to a pronounced increase in activity in the GDP/GTP exchange assay (IC50 values in the nanomolar range against KRasG12D and low micromolar range against KRasG12V). Glide docking and molecular dynamics simulations revealed a previously uncharacterized allosteric binding pocket at the KRas4Ballo:SOS interface, in which dimethoxyphthalate derivatives form stable complexes (up to -12.2 kcal/mol). The key binding motif involves chelation of the Mg2+ ion by a phthalate residue, while the triterpene scaffold aligns along the Rasallo-SOSRem interface. This interaction disrupts the Tyr64Ras-SOSRem contact, which is essential for stabilizing the catalytically active SOS conformation, thereby preventing GDP/GTP exchange. Despite their limited drug-likeness, phthalate-modified triterpenes represent a promising starting point for the development of structurally distinct, allosteric pan-Ras inhibitors capable of targeting even the pharmacologically challenging KRasG12V mutant.
